Regulatory T cells (Tregs) participate in the negative regulation of inflammatory reactions by suppressing effector cells. In a number autoimmune disorders, suppressive function and/or Tregs is compromised. The lack active functioning can be restored with adoptive transfer expanded ex vivo autologous Tregs. our study, we traced differentiation and maturation CD4+CD25+FoxP3+CD127low over 7 days cultivation from initial CD4+ under conditions. resulting cell population (eTregs) demonstrated immune profile an increased capacity to suppress proliferation target expression FoxP3 gene was upregulated within time expansion associated gradual demethylation promotor region cell-specific region. Real-time RT-PCR analysis revealed changes genes involved cycle regulation. addition FOXP3, displayed elevated mRNA levels Ikaros zinc finger transcription factors main telomerase catalytic subunit hTERT. Alternative splicing FoxP3, hTERT IKZF family members eTreg maturation. Our data indicate that eTregs develop Treg-specific phenotype functional activity.
